EPHA4
EPHA4全稱為促紅細(xì)胞生成素肝細(xì)胞受體A4，屬于Eph受體酪氨酸激酶家族成員，常被稱為EPH受體A4或酪氨酸蛋白激酶受體Tyro3。EPHA4通過與其配體ephrin相互作用，在神經(jīng)系統(tǒng)發(fā)育中發(fā)揮關(guān)鍵作用，尤其參與調(diào)控軸突導(dǎo)向和神經(jīng)元連接的形成。EPHA4異常表達(dá)與肌萎縮側(cè)索硬化癥（ALS）密切相關(guān)，其活性降低可能加速運(yùn)動(dòng)神經(jīng)元退化進(jìn)程，相關(guān)基因療法已在動(dòng)物模型中顯示出延長生存期的效果。在腫瘤領(lǐng)域，EPHA4在結(jié)直腸癌、膠質(zhì)母細(xì)胞瘤等惡性腫瘤中呈現(xiàn)高表達(dá)特征，通過影響腫瘤細(xì)胞侵襲轉(zhuǎn)移能力促進(jìn)疾病進(jìn)展。針對(duì)該靶點(diǎn)的藥物開發(fā)聚焦于小分子抑制劑和單克隆抗體，其中選擇性抑制劑ALW-II-41-27在臨床前研究中展現(xiàn)出抑制腫瘤轉(zhuǎn)移的潛力，而基于CRISPR技術(shù)的基因編輯療法正探索其在神經(jīng)退行性疾病中的應(yīng)用。
